Ziihera shows overall survival benefit over trastuzumab in stomach cancer
Jazz Pharmaceuticals reported that a second overall survival (OS) win in the phase 3 Herizon-GEA-01 trial showed Ziihera plus chemotherapy significantly improved OS versus trastuzumab plus chemotherapy, strengthening the bispecific antibody's case as the new HER2 backbone in first-line HER2-positive gastroesophageal adenocarcinoma. An earlier interim readout also showed the Ziihera-Tevimbra-chemo triplet produced a 28% OS improvement versus trastuzumab-chemo. The FDA approved Ziihera for first-line GEA on Aug. 25 with a black box warning for life-threatening diarrhea; the triplet label covers IHC 3+ and IHC 2+/ISH+ tumors, while the doublet is limited to IHC 3+.
