Developing joint cells linked to later rheumatoid arthritis vulnerability
Scientists at the Kennedy Institute used single-cell sequencing, advanced image analysis and high-resolution 3D X-ray scanning to map developing human finger joints and identify structural and cellular differences present before birth. They found that proximal interphalangeal (PIP) joints had more synovial tissue and higher numbers of PI16-positive fibroblasts than distal interphalangeal (DIP) joints, and that PI16+ fibroblasts show distinct responses to inflammatory signals. The study, published in Nature Immunology, suggests that developmental tissue features may help determine joint susceptibility to rheumatoid arthritis and could redirect research into developmental signals and fibroblast biology.
